Overview

Peptidyl-prolyl cis/trans isomerase (PPIase) that binds to and isomerizes specific phosphorylated Ser/Thr-Pro (pSer/Thr-Pro) motifs in a subset of proteins, resulting in conformational changes in the proteins (PubMed:21497122, PubMed:22033920). Displays a preference for an acidic residue N-terminal to the isomerized proline bond. Regulates mitosis presumably by interacting with NIMA and attenuating its mitosis-promoting activity. Down-regulates kinase activity of BTK (PubMed:16644721). Can transactivate multiple oncogenes and induce centrosome amplification, chromosome instability and cell transformation. Required for the efficient dephosphorylation and recycling of RAF1 after mitogen activation (PubMed:15664191). Binds and targets PML and BCL6 for degradation in a phosphorylation-dependent manner (PubMed:17828269). Acts as a regulator of JNK cascade by binding to phosphorylated FBXW7, disrupting FBXW7 dimerization and promoting FBXW7 autoubiquitination and degradation: degradation of FBXW7 leads to subsequent stabilization of JUN (PubMed:22608923).

Molecular Function

GTPase activating protein binding; mitogen-activated protein kinase kinase binding; peptidyl-prolyl cis-trans isomerase activity; phosphoserine binding; phosphothreonine binding

Subcellular Location

Nucleus; Nucleus speckle; Cytoplasm

Expression

The phosphorylated form at Ser-71 is expressed in normal breast tissue cells but not in breast cancer cells.

Structure

Interacts with STIL. Interacts with KIF20B. Interacts with NEK6. Interacts (via WW domain) with PRKX. Interacts with BTK. Interacts (via PpiC domain) with DAPK1. Interacts with the phosphorylated form of RAF1. Interacts (via WW domain) with ATCAY; upon NGF stimulation. Interacts with PML (isoform PML-4) and BCL-6. Interacts with FBXW7, disrupting FBXW7 dimerization and promoting FBXW7 autoubiquitination and degradation (PubMed:22608923).

Overview

May function as calcium sensor and contribute to cellular calcium signaling. In a calcium-dependent manner, functions by interacting with other proteins, such as EZR and PPP5C, and indirectly plays a role in physiological processes like the formation of microvilli in epithelial cells. May stimulate cell proliferation in an autocrine manner via activation of the receptor for activated glycation end products (RAGE).

Molecular Function

Calcium-dependent protein binding, calcium ion binding, magnesium ion binding

Subcellular Location

Nucleus, cytoplasm, Cell projection > microvillus membrane

Expression

Detected in all of the tissues except brain, testis and small intestine, expression level is higher in placenta, heart, lung, skeletal muscle, spleen and leukocyte. Up-regulated in various pancreatic ductal adenocarcinomas and pancreatic intraepithelial n

Structure

Homodimer and heterodimer with S100A1. Interacts with S100PBP and S100Z. Interacts with CACYBP in a calcium-dependent manner. Dimeric form binds to and activates EZR/Ezrin by unmasking its F-actin binding sites. Interacts with PPP5C (via TPR repeats); the
