Do you have any suggestions about the PDE3b antibody? It gives bands about 100 for different concentrations I used. I loaded 20 micrograms of cortex and cerebellum lysates and incubated the nitrocellulose membranes with the PDE1(a,b,c) and PDE3b antibodies.
1. Thank you for your inquiry about PD3B-101AP,
  I looked at the gel images you provided and am curious why you are using a high percentage or gradient gel for a protein that is above 100kDa. There are two issues that I can think of that may be affecting the results. The high percentage acrylamide may be reducing the efficiency of high MW proteins onto nitrocellulose membrane. The cortex is not the regions where we see PDE3B in high density, so it may be possible that the levels of PDE3B is already below the detection limit here. Second issue may be due to proteolysis. PDE3B is cleaved off by endogenous proteases if their activity is not checked properly. In order to curb this prepare the homogenates and samples in the presence of Fabgennix Protease Inhibitor Cocktail before the SDS-PAGE. Use PC-PDE3B to identify bands and validate the system.
